Ih and GABAB in CA1 afterhyperpolarizations Contribution of Ih and GABAB to synaptically-induced afterhyperpolarizations in CA1: a brake on the NMDA response

چکیده
CA1 pyramidal cells receive two major excitatory inputs; the perforant path (PP) terminates in the most distal dendrites whereas the Schaffer collateral (SC) terminates more proximally. We have examined the mechanism of the afterhyperpolarization (AHP) that follows single subthreshold EPSPs in these inputs. The AHPs were not reduced by a GABA A antagonist or by agents that block Ca 2+ entry. Application of the I h blocker, ZD7288 partially blocked the AHP in the PP; the substantial remaining component was blocked by 2-hydroxysaclofen, a GABA B antagonist. In contrast, the AHP in the SC depends nearly completely on I h , with almost no GABA B component. Thus postsynaptic GABA B receptors appear to be preferentially involved at distal synapses, consistent with the spatial distribution of GABA B receptors and GIRK channels. GABA B does however play a role at proximal synapses through presynaptic suppression of glutamate release, a mechanism that is much weaker at distal synapses. Experiments were conducted to explore the functional role of the AHP in the PP, which has a higher NMDA/AMPA ratio than the SC. Blockade of the AHP converted a response that had a small NMDA component to one that had a large component. These results indicate that the I h and postsynaptic GABA B conductances act as a brake on distally generated NMDA responses.

متن کاملThe effect of GABAB receptors in CA1 region of hippocampus on morphine tolerance in female Wistar race rats by conditioned place preference
Background and Aim: The hippocampus is a region of the brain that plays an important role in learning activities, including conditioned place preference (CPP). This area is widely used as the target of the studies on the reinforcing effects of the drugs and narcotics. Considering the wide distribution of GABAB receptors in the hippocampus CA1 region, these receptors are likely to play an import...
متن کاملHigh intracellular Cl- concentrations depress G-protein-modulated ionic conductances.
Numerous G-protein-modulated ionic conductances are present in central neurons and play major roles in regulating neuronal excitability. Accordingly, endogenous factors that alter the operation of these conductances may have profound effects on neuronal function. We now report that several G-protein-modulated ionic conductances in hippocampal neurons are very much altered when Cl- is the predom...
متن کاملGABAB-receptor-mediated inhibition of the N-methyl-D-aspartate component of synaptic transmission in the rat hippocampus.
GABA receptor regulation of NMDA-receptor-mediated synaptic responses was studied in area CA1 of the rat hippocampus using extracellular and intracellular recording techniques. Picrotoxin (PTX) was used to suppress GABAA inhibition and 6,7-dinitroquinoxaline-2,3-dione (DNQX) was used to suppress non-NMDA receptor-mediated responses. In this manner, we were able to avoid the complicating factors...
متن کاملThe role of GABAB receptors in imipramine-induced antinociception in experimental model of neuropathic pain
This study was designed to investigate the role of GABAB receptor agents on imipramine-induced antinociception in ligated and non-ligated mice using hot-plate test. The data showed that different doses of morphine (3, 6, and 9 mg/kg) induced a dose-dependent antinociception in ligated and non-ligated mice. However, the opioid response was decreased in ligated animals. Intracerebroventricular (i...
